Overview

KY19382 (5,6-dichloroindirubin-3'-methoxime) is a synthetic analogue of indirubin-3'-monoxime developed at Yonsei University. In the published literature it is characterised as an activator of Wnt/beta-catenin signalling that works by inhibiting the interaction between CXXC-type zinc finger protein 5 (CXXC5), a negative feedback regulator of the pathway, and dishevelled (Dvl)1,2.

It was first reported in a 2019 study of growth plate senescence and longitudinal bone growth in mice1, and has since been studied in hair follicle, cutaneous wound, diabetic wound and skin carcinoma models2,3,4,5,6. Most of the work comes from the originating group, whose authors disclose that the compounds are licensed to a company they are affiliated with1,2,6.

Mechanism of action

As described in the research literature

As described in the research literature, an in vitro screen for compounds that disrupt the CXXC5-DVL interaction identified several indirubin analogues, from which KY19382 was developed as a functionally improved derivative1. In human dermal papilla cells it activated Wnt/beta-catenin signalling and increased alkaline phosphatase and the proliferation marker PCNA2, and in human keratinocytes and dermal fibroblasts it increased beta-catenin, Keratin 14, PCNA, Collagen I and alpha-smooth muscle actin4.

An independent group studying diabetic wounds reported that blocking CXXC5 stabilised Wnt/beta-catenin signalling by reducing binding of the repressor CTBP1 to beta-catenin, with downstream effects on VEGFA/VEGFR2 and NF-kappaB signalling in endothelial cells5. In cutaneous squamous cell carcinoma cells, KY19382 was reported to inhibit Wnt/beta-catenin signalling in association with suppressed CDK1 expression, showing that its effect on the pathway depends on cellular context6.

Research areas

What published studies have examined

Growth plate and longitudinal bone growth

Cxxc5 knockout mice were used to examine the role of CXXC5 in growth plate senescence, and KY19382 was studied for its effect on tibial length and growth plate activity in adolescent mice1.

Hair follicle models

Human dermal papilla cells, ex vivo human hair follicles and mouse vibrissae, and mouse hair regrowth, wound-induced hair follicle neogenesis and hair patch assays were used to study KY193822. A follow-up study examined hair regrowth and follicle neogenesis in db/db and diet plus streptozotocin-induced diabetic mice, and human hair follicles cultured in high glucose3.

Wound healing models

Keratinocyte and fibroblast migration assays and a murine cutaneous wound model examined re-epithelialisation, collagen deposition and stem cell markers4. An independent study examined diabetic mouse wounds, dermal fibroblasts and umbilical vein endothelial cells under high glucose5.

Skin carcinoma models

Human cutaneous squamous cell carcinoma cells and a two-stage mouse skin carcinogenesis model were used to examine KY19382 and the related compound KY19334, with CDK1 knockdown used to validate the pathway6.
